Эндокринные разрушители (endocrine disruptors) оказывают негативное влияние на функционирование эндокринных органов, приводя к развитию нежелательных реакций. Эндокринная токсичность требует особого внимания при проведении доклинических исследований потенциальных лекарственных средств.</p></sec><sec><title>Цель</title><p>Цель. Анализ международных рекомендаций и подходов к оценке рисков эндокринной токсичности лекарственных препаратов.</p></sec><sec><title>Обсуждение</title><p>Обсуждение. Проведен анализ документов, которые являются методической основой для выявления и отнесения соединений к классу эндокринных разрушителей — Руководства Организации экономического сотрудничества и развития (OECD) по оценке токсичности химических веществ для эндокринной системы (2018 г.), Руководства для отрасли по доклинической оценке токсичности лекарственных средств для эндокринной системы (Управление по контролю за качеством продуктов питания и лекарственных средств США, 2015 г.) и Руководящего документа Европейского союза, разработанного в соответствии с Регламентом о биоцидных продуктах (ЕС) № 528/2012 и Регламентом о средствах защиты растений (ЕС) № 1107/2009. Предложен алгоритм оценки эндокринной токсичности, который включает сбор всей доступной информации о тестируемом соединении: данные литературы, ранее полученные экспериментальные данные, в том числе об острой и субхронической токсичности, а также прогнозные оценки, полученные с помощью in silico методов. Основное внимание следует уделить результатам проведения стандартных доклинических исследований хронической токсичности, которые в большинстве случаев позволяют выявить побочные эффекты, связанные с эндокринной системой. Ключевыми критическими точками проявления эндокринотоксичности являются изменение массы и гистопатология основных эндокринных органов (надпочечники, семенники, эпидидимисы, яичники, щитовидная железа), влияние на эструс, репродуктивная токсичность и трансплацентарное действие. Тщательная оценка полученных данных позволит сделать вывод о наличии или отсутствии неблагоприятной эндокринной активности и о необходимости проведения дополнительных исследований.</p></sec><sec><title>Выводы</title><p>Выводы. Валидированные in vivo и in vitro тесты, описанные в Руководстве OECD, позволяют в зависимости от выявленных токсических эффектов охарактеризовать важнейшие механизмы эндокринной токсичности: эстрогенный, андрогенный, тиреоидный и влияние на стероидогенез. Использование такого подхода позволит исследователям выявить потенциальные эффекты эндокринной токсичности на ранних стадиях разработки лекарственных средств и оптимизировать необходимый объем исследований.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Scientific relevance</title><p>Scientific relevance. Endocrine disruptors affect the functioning of endocrine organs, which leads to adverse drug reactions. Endocrine toxicity requires special attention in preclinical studies of candidate medicinal products.</p></sec><sec><title>Aim</title><p>Aim. The study aimed to review international guidelines and approaches to assessing the risk of endocrine toxicity associated with medicinal products.</p></sec><sec><title>Discussion</title><p>Discussion. This review covers documents that provide a methodological framework for identifying and classifying a chemical compound as an endocrine disruptor. These documents include the following: Revised Guidance Document 150 on Standardised Test Guidelines for Evaluating Chemicals for Endocrine Disruption (Organisation for Economic Cooperation and Development, 2018), Nonclinical Evaluation of Endocrine-Related Drug Toxicity (Food and Drug Administration, 2015), and Guidance for the Identification of Endocrine Disruptors in the Context of Regulations (EU) No. 528/2012 and (EC) No. 1107/2009. The proposed algorithm for endocrine toxicity assessment involves collecting all available data on the test compound, such as the literature and previously obtained experimental data, including acute and subchronic toxicity data, and in silico predictions. Particular attention should be paid to the standard battery of preclinical chronic toxicity studies, which can identify most side effects associated with the endocrine system. The main endpoints for endocrine toxicity include changes in the mass and histopathology of the major endocrine organs (adrenal glands, testes, epididymides, ovaries, and the thyroid gland), oestrous cycle effects, reproductive toxicity, and transplacental action. A thorough assessment of the data obtained provides for the determination of unfavourable endocrine activity that requires further studies.</p></sec><sec><title>Conclusions</title><p>Conclusions. The OECD guidelines offer a set of validated in vivo and in vitro tests that characterise the most important mechanisms of endocrine toxicity (oestrogen, androgen, thyroid, and steroidogenic endocrine pathways) by identified toxic effects.
